Right now? Tough one. With the last two major transfers belonging to Real , you might have to side with them.

But Man U/Barca/Real all have good cases. Definitely one of those 3 though.

Barcelona and Manchester United are the two leaders at the moment, that may change with Real's spending spree, but overall, when you include history, Real are for me the biggest club.

I'd also go out on a limb to say I consider Liverpool as a bigger club than Man Utd when you consider honours and history.

Just remember that La Liga has been dominated by Madrid and Barca for years now (only 9 clubs have ever won it ever, and since the war it's been almost as bad as Scotland), so I don't think you can say it's got a history of being such a competitive league. Also, of the 9 European Cups, 5 were the first five years in a row, so I don't think it shows them being as dominating over the years as the number 9 makes it sound (although of course they still have 4 others!). And remember, if not for Munich, United would have had a big shout for one of those, and maybe more.
However, overall, I definitely think Madrid are the biggest club.
I also agree that Liverpool are bigger then United history-wise. The major point which makes me say this is the utter dominance United have had since the 90's, which I don't believe Liverpool ever had for such a period. This may round a bit illogical, but to my mind a short period of utter dominance - like United the last decade and a half, and similarly Madrid in the early years in Europe - is nowhere near as good as a longer period with less sheer dominance. At least when considering which club is bigger from a historical perspective.

I disagree on your diminishment of the competitivity of La Liga, the standard of player, team and football there is fantastic.

Also, yes, they won the initial five ECCs, but they were an awesome team, the 7-3 final at Hampden is often referred to as one of the great performances by a club team. Di Stefano and Puskas were arguably the two best players in the world at that moment in time, the format was the same as the 1988 ECC competition, it wasn't easier to win it or anything like that, although arguably the new CL format is harder to win.

For me it's Real > Barca > AC Milan > Liverpool > Man Utd.

I wasn't diminishing those 5, just saying that it was all in just 5 years. It's not like they won 9 spread out over the entire 50 years of european competition, similar to my last point about United recently. Concerning the competitiveness of the league, from what I've read about the history of la liga it certainly was not as competitive outside the top teams as the football league was in england in those days.
