The first set was tricky for the former world number one, who was broken twice by his Argentinian opponent. Murray cut a frustrated figure, unable to convert the opportunities he had against Londero's serve.
But the Brit improved notably in the second set, breaking Londero in the Argentine's first and last service games to take the set by a commanding 6-2 scoreline.
The third set was close, with the pair exchanging breaks in the fourth and fifth games. But Murray made the decisive move in game six on route to victory – his first ATP Masters 1000 win since May 2017.
Speaking after the match, Murray said:
"I got better as the match went on – I had to start going for my shots a bit more."
Murray will face world number 12 Fabio Fognini in the next round.
